They read the script, turn each role into a breakdown of the character, gather candidates from the talent pool, and run auditions, chemistry reads, and callbacks. From this they shape the cast, the performers who fill each role. The need arrives from the screenwriter's script and the director's vision, and the chosen actors carry the parts forward on set. The casting director sets the human core the whole film is built around.
